CONCLUSIONS Our main result is that continuum electrostatics using a rigid protein structure cannot provide a consistent description of all the available data on the GA channel. Surprisingly, the use of a dielectric constant of 80 in the channel appears to give the best results in our continuum electrostatics, despite the fact that from microscopic considerations such a high value seems unreasonable in the narrow GA channel. Although slight changes in the protein structure may alter the energy profiles, the experimental data limit the amount of flexibility allowed, and it seems unlikely that all the problems encountered could be overcome using a different or flexible protein.
