2.2.2 DNA isolation using CTAB method Total genomic DNA was isolated using a modified hexadecyltrimethylammonium bromide CTAB ; method based on Saghai-Maroof et al. 1984 ; . Preferably young Clivia leaves were freeze-dried using the FreezeMobile II apparatus ; and ground to a fine powder in the presence of silica gel using a mortar and pestle. A volume of 750 l CTAB buffer [100 mM tris hydroxymethyl ; aminomethane Tris ; , pH 8.0; 20 mM ethylenediaminetetraacetate EDTA ; , pH 8.0; 1.4 M NaCl; 2% w v ; CTAB and 0.2% v v ; -mercaptho-ethanol] was added to approximately 250 l fine leaf powder in a 1.5 ml microfuge tube and incubated at 65oC for one hour. A volume of 500 l chloroform: isoamylalcohol [24: 1 v v ; was added to the suspension. Phases were separated by centrifugation at 12 000 g for 3 minutes. DNA was precipitated from the aqueous phase with 0.66 volumes 2-isopropanol at room temperature for 20 minutes. Centrifugation followed at 12 000 g for 10 minutes. The supernatant was discarded and tubes drained upside down. The precipitate was washed at room temperature with 500 l 70% v v ; ethanol for 20 minutes followed by centrifugation at 12 000 g for 10 minutes. The pellet was air-dried for one hour and resuspended in TE buffer 10 mM Tris.Cl, pH 8.0; 1 mM EDTA, pH 8.0 ; . Overnight incubation followed at 4oC. DNasefree RNase 0.1g l ; was added to samples and incubation at 37oC followed for 2 hours. Ammonium acetate 0.75 M ; and an equal volume of chloroform: isoamylalcohol [24: 1 v v ; were added to the samples. DNA was precipitated from the aqueous layer with two volumes of ice-cold absolute ethanol followed by overnight incubation at -20C. DNA was recovered by centrifugation at 12 000 g for 15 minutes and washed twice with cold 70% v v ; ethanol by centrifugation of 10 minutes each time. The pellet was air-dried and resuspended in TE buffer. DNA quantity and quality were estimated using an UV spectrophotometer by measuring.

Since the differences in concentration of blood and urine were maintained. They concluded that the cells of the renal epithelium were capable of secreting into the urine whatever molecular species was present in the blood in abnormally high amount. The authors proposed finally that the organs present in the renal sac were capable of reabsorption. When they injected glucose or urea into the renal sacs and ligated the urinary papillae, the concentration of the injected substance remained high in the urine for a few hours but was reduced to a mere trace 24 hr. after the beginning of the experiment. They did not attempt to identify the specific structures concerned in the reabsorption and did not propose a mechanism by which this was accomplished. Bruni 1937 ; reported a somewhat similar type of study employing the injection of dyes intramuscularly. He followed the changes in concentration of dye in the urine, but not in the blood. He reported that different dyes are eliminated at different rates and that they are capable of inducing different degrees of polyuria. He concluded that the coloured materials were eliminated by a process of filtration. Although important contributions have been made by these investigators, some of their conclusions are contradictory. In addition, the techniques employed did not permit evaluation of the dynamics of the processes going on in the animal. It has been the purpose of this investigation to try to establish a few of the mechanisms functioning in urine formation in the octopus under conditions as physiological as possible. The technique adopted was to follow for extended periods of time the concentration of specific agents in samples of blood, urine and pericardial fluid taken serially before, during and after the administration of metabolic poisons.

The National Provider Identifier NPI ; is a unique 10-digit code required by HIPAA that will eventually replace the various identifiers that physicians and other providers currently use when rendering an electronic transaction to a public or private payer. Covered healthcare providers, including health plans, healthcare clearinghouses and those physicians who transmit health information in electronic form in connection with a HIPAA standard transaction, will be required to begin using their NPI by May 23, 2007 for electronic claims and other HIPAA standard transactions. Although the May 23, 2007 deadline marks Medicare's transition to the NPI, other health plans may require contracted physicians to utilize their NPI earlier. Therefore, it is best to confirm the use of the NPI with each carrier with which you work. The NPI should not be used prior to this date unless a health plan specifically instructs you to do so. For more information and to apply for a NPI electronically or by paper CMS-10114 form ; , visit the National Plan and Provider Enumeration System Web site at s: nppes.cms.hhs.gov NPPES.

QUEER SEXUALITIES: DIS LOCATING OUR "CHAINS OF SIGNIFICATION." Sponsor: Gay Lesbian Bisexual Transgendered Communication Studies Division Chair: Naida Zukic, Southern Illinois University, Carbondale Participants: Naida Zukic, Southern Illinois University, Carbondale Cheryl L. Nicholas, Southern Illinois University, Carbondale Raquel Monroe, University of California, Los Angeles Makoto Saito, University of Massachusetts, Amherst Ahmet Atay, Southern Illinois University, Carbondale Respondent: Shane Moreman, California State University Fresno In this panel we aim to politicize queer formations and to intervene as agents of change by challenging the narrowly Eurocentric representations of queer sexualities. We set out to examine queer experiences by looking "beyond the borders of North America and Europe to more seriously address how queer identities and queer cultural formations have taken shape and operate elsewhere" Spurlin 185-6 ; . Consequently, the panel interrogates the theoretical and methodological framings of queer identities in contexts where markers of difference interweave with various institutionalized structures of meaning. Experiences of queerness are articulated vis--vis the questions of agency, discourses of globalization, alienation, and commodification. Importantly then, this panel underscores queerness as embraced critical methodology that affords acute insights and understandings of contested dynamics of sexual identities, in particular, and the discourses about structures that determine and constrain such identities, in general. 20255 9: 30 to 10: 45 Convention Center 2nd Level Room 204 B. Characterization of Hyaluronun-associated Serum Proteins sequences. The sequences corresponded to the sequence of the heavy chain 1 HC1 ; of IT1 from position 353. Considering their apparent molecular masses the 37-kDa chain corresponds to approximately 340 amino acid residues ; , and the HA-binding domain in the HC1 chain must be localized between position 353 and near the COOH-terminal end, position 638. Consistent with this suggestion, a computerassisted analysis 37 ; identified an amphipathic CY helical structure residues 508-563 ; that may bind to HA.
